Özet

Background The multi-country Nucala Effectiveness Study (NEST) assessed mepolizumab (MEPO) effectiveness in patients with severe asthma with an eosinophilic phenotype (SAEP; Al-Lehebi, R.O. et al. Adv Ther 2024;41:4008–31). Aim Summarise data on clinically significant exacerbations (CSE) and oral corticosteroid (OCS) use in NEST Türkiye cohort. Methods This bi-directional study assessed 12 months pre- and post-MEPO initiation data. Primary endpoint: incidence rate ratio (IRR) of CSEs (asthma worsening requiring systemic corticosteroid use for ≥3 days and/or emergency room visit and/or hospital admission). Key secondary endpoint: change in maintenance OCS use. Results The cohort comprised 150 patients (mean age: 48 years; 76% female). MEPO exposure increased the number of patients with no CSEs (122 [81.9%]) versus pre-MEPO (36 [25.7%]). The proportion of patients with ≥1 CSE was lower post-MEPO (18.1%) than pre-MEPO (74.3%). The mean (95% confidence interval [CI]) number of CSEs/patient in pre- and post-MEPO periods was 2.9 (2.4–3.5) and 0.3 (0.2–0.4), respectively. CSEs reduced by 91% post-MEPO versus pre-MEPO (IRR [95%CI]: 0.09 [0.06–0.14]; p<0.001). Maintenance OCS use decreased from 125 (83.3%) patients pre-MEPO to 32 (21.3%) post-MEPO (Figure). Conclusions MEPO significantly reduced CSE frequency and OCS use in patients with SAEP supporting its clinical use in SAEP maintenance. erj;66/suppl_69/PA320/F1 F1 F1
