Summary
Parkin is an E3 ubiquitin-protein ligase that plays an important role in various biological processes such as cell cycle, cell proliferation, apoptosis, metastasis, and mitophagy. Parkin interacts with activators Cdc20 and Cdh1 and it is involved in the degradation of mitotic regulatory proteins. It has been determined that HN1 also has effects on the proteins involved in the mitotic phase and on Cdh1/Cdc20. The absence of HN1 and Parkin in cells has been implicated in high levels of target proteins, mitosis errors, genome instability, and the development of the tumorigenic phenotype. Therefore, it was thought that the contribution of HN1 to mitotic regulation might be Parkin-mediated, and it was aimed to investigate the deficiency in the literature. In this project, as a continuation of our previous studies, we have elucidated the effect of HN1 and also Parkin on the cell cycle in the SH-SY5Y neuroblastoma cell line. Data from the project show that HN1 can influence Parkin expression and the cell cycle regulation by such mechanisms, which needs to be elucidated in further studies. Since the data obtained is related to the cell cycle and mitosis control, it can be published in high-impact factor journals. In addition, this project is contributed to the training of graduate students who are equipped with cancer biology.
