Summary

Sars-CoV-2 virus causes COVID-19 disease with a high mortality rate and symptoms such as severe acute respiratory failure. There is no effective treatment for this disease yet. However, it has been proven that 3 chymotrypsin-like protease (3Clpro) enzymes, which have a key role in Sars-CoV-2 virus replication, can be inhibited by some natural compounds in flavonoid structure. Flavonoids interact with the active site of the enzyme and provide enzyme inhibition. In this project, it is planned to determine the 3Clpro inhibitory concentrations of flavonoid molecules and to obtain the most accurate extracts that can be effective in enzyme inhibition of natural plant extracts containing these molecules at high levels. The aim of our project is the effect of extracts obtained from the root of licorice (Glycyrrhiza Glabra L.) plant, which is known to be rich in flavonoid molecules, the stem of oudi turkey (Aquilaria Agallocha) and woad (Isatis tinctorial) plants by different extraction methods, on the inhibition of 3Clpro enzyme and the effects of Sars-CoV- To investigate its possible role in the treatment of 2 virus infections. In our study, it is a completely original research since the phenolic content of these natural products was determined for the first time by LC MS/MS and the 3Clpro enzyme inhibitory effect of these natural product extracts has been studied for the first time.

Antioxidant activity (DPPH) of Lsatis tinctorial plant was low in water-soluble extracts ( W-RT-6 h, W-RT-12h, W-RT-24h,W-40°C-24h,US-5min,MW- 15 sec)and was high in all other extracts. Ferulic acid, epicatechin and delphidin are the major phenolic contents in all extracts.

Amentoflavone, campherol, chlorogenic acid and caffeic acid content were found to be the highest in Aquilaria Agallocha plant dissolved in 80% ethanol, and the antioxidant activity of this extract (DPPH, TEAC) was found to be higher than the others.

When Glycyrrhiza Glabra L extracts were evaluated in terms of antioxidant parameters, the highest TEAC, FRAP and DPPH levels were obtained, especially in ethanol extracts.

The results of our study showed that the antioxidant and anti-inflammatory effects of natural products vary depending on the solvent and temperature. Lsatis tinctorial, Aquilaria Agallocha and Glycyrrhiza Glabra L plants have the potential to exert therapeutic effects against the SARS COV-2 virus when properly prepared
