Identification of the Aspergillus MF44 strain isolated from a marine sponge, Axinella sp, as a new species, Aspergillus marinensis nov.
Project no. 23669 · Status: Kapanmış (Yayın bekleniyor) · 2022
Summary

Within the scope of the project, complementary phenotypic and genotypic data was obtained for the identification of the MF44 coded filamentous fungus strain isolated within the scope of a TUBITAK project (Project no: 109S361) on the discovery of bioactive metabolites from marine microorganisms. MF44 strain was isolated from Axinella sp. collected from the coast of Mersin, and as a result of the analysis carried out for its identification, it was determined that it could be a new Aspergillus species. In addition to the previously performed cultural and microscopic tests and ITS sequence analysis, ekstrolit analysis and whole genome sequence analysis were performed. It was determined that the extract obtained from the Aspergillus sp. MF44 strain, whose extrolite analyses were carried out using the MS/MS method, contained 5 different components. All of these components have various activities and are compounds previously identified from various Aspergillus species. Whole genome analysis was performed with 100x coverage and a total of 1700 contigs were obtained. The ITS 1 gene of MF44 was identified in contig number 30. The ITS1 gene region of the isolate numbered MF44 was compared with the 3401 different ITS1 gene region sequences belonging to the Aspergillus genus in NCBI using the MAFFT program. Aspergillus sp. strain MF44 showed a distinct clustering. The new data obtained also supports that the MF44 strain is a new species. The species Aspergillus marinensis nov. it is planned to be named and sent for publication as soon as possible.
