INVESTIGATION OF THE PRESENCE OF RHIZOXIN PRODUCER ENDOPHUNGAL BACTERIA IN THE MEMBERS OF RHISOPUS GENUS INSULATED FROM THE SOIL SAMPLES FROM THE AGRICULTURAL LANDS OF THE DISTRICTS OF IZMIR
Project no. 560 · Status: Kapanmış · 2017
Summary
In our study, isolation of rhizoxin-producing endofungal bacteria that have an an anti-mitotic activity from Rhizopus species, which is a prevalent plant pathogen in Izmir Provinces’ agricultural areas where agricultural output and crop yields are considerably high. In the study, 237 Rhizopus spp. isolated from soil samples taken from 237 different agricultural lands. Rhizopus species were identified phenotypically. Phenotypic diagnosis has also been confirmed by molecular studies using two different sets of primers targeting two different gene regions. The presence of endofungal bacteria was investigated in all 237 isolates of Rhizopus spp. Endofungal bacteria were isolated from 20 Rhizopus spp. Bacteria were identified as molecular by 2 different primer sets. According to this; Alcaligenes faecalis (11), Bacillus circulans (1), Bacillus thuringiensis (3), Bacillus cereus (1), Klebsiella pneumoniae (2), Leucobacter aridicolis (1) and Serratia marcescens (1) species were isolated as endofungal. Endofungal isolation of Alcaligenes faecalis, Bacillus circulans, Bacillus thuringiensis, Bacillus cereus and Leucobacter aridicolis species was made for the first time. Our study contributed to the diversity of endofungal bacteria. In our study, the presence of ketosentase gene was found in Alcaligenes faecalis (B 88, B93 and B180 coded isolates), Bacillus cereus (B67 coded isolate), Bacillus thuringiensis (B40, B99 and B152 coded isolates). The bacterial isolates with the ketosentase gene were investigated for rhizoxin production separation-purification, spectroscopic analysis and in B180 coded isolate (Alcaligenes faecalis) rhizoxin was identified. Our study is important since it is the first study showing rhizoxin production in a species (Alcaligenes faecalis) other than Burkholderia rhizoxinica.
