Evaluation of the cytotoxic, proapoptotic and epigenetic effects of thyme oil obtained from Origanum onites L. on healthy gastric cell line and gastric cancer cell line
Project no. 22453 · Status: Kapanmış · 2021
Summary

Origanum onites L., known as Turkish thyme, is used in many areas throughout the country. Especially in the Aegean region, it is believed that it is good for many diseases and ailments, and it is consumed orally, especially for gastric and dyspeptic complaints. With the widespread use of essential fatty acids in the field of pharmacology, this belief has also attracted the attention of the scientific world. Many studies have been conducted on the content and biological effects of Origanum onites L. Most of the remarkable studies are on the antimicrobial effect of the molecules contained in this plant. There is little research on its anticancer effect and these studies are limited to its antiproliferative, antimigratory and pro-apoptotic effects on cancer cells.

Thyme oil is mostly used topically and orally in our country. When taken orally, the first cells to encounter the molecules it contains are the epithelial cells of the digestive tract. Based on this logic, after analyzing the content of Origanum Onites L. extract and determining its IC50 value, its cytotoxic, pro-apoptotic and epigenetic effects on healthy and cancerous human gastric epithelial cell lines, which constitute a large part of the original value of our project, were evaluated. It has cytotoxic and pro-apoptotic effects on cancerous cells, increases the expression of many tumor suppressor genes involved in the Wnt/β-catenin signaling pathway in cancerous and healthy cells, decreases the expression of oncogenes, and also modifies the methylation pattern of oncogenes and tumor suppressor genes in healthy and gastric cancer cells. In line with the results of the research, it is thought that the potential of this rare plant in our geography to be used in the production of anticancer drugs will pave the way.
