THE DETERMINATION OF BIOACTIVITIES OF THE SMOOTH NEWT (LISSOTRITON VULGARIS) SKIN SECRETION AND THE EFFECTS ON SOME CANCER CELLS

Project no. 421 · Status: Kapanmış · 2016

Summary

In this thesis work, skin secretion of the smooth newt (Lissotriton vulgaris) was studied, which is distributed in İzmir province. In this context, a detailed field study has been organized so that the target can be found and skin secretion can be obtained.Examples of cutaneous aggregates collected from individuals with small intense electrical currents have been assembled and lyophilized for use in bioactivity tests.Protein content, SDS-PAGE protein profile, antimicrobial activity, healthy cell lines and anticarcinogenic effects on cancer cell lines were determined for skin secretions from individuals. In the thesis study hemolytic effects on the blood cells of the skin epidemic were also investigated. The total protein content of L. vulgaris skin secretion was determined to be 1775 µg/ml.The resulting polyacrylamide gel electrophoresis showed that the resulting skin secretion contained 18 protein fractions in the 9-265 kDa range. As a result of MIC test performed, crude skin secretion showed effects on E. coli and C. albicans at 7.8µg/ml.Skin secretion as a result of MBC and MFC tests showed an effect on E. coli and C. albicans at 31.25 µg/ml. Result of the cytotoxicity tests, the crude skin secretions showed effect on MDA-MB-231 (mammary gland) and PC-3 (prostate) cancer cell lines, respectively, with IC50 values of 1.58 and 4.99 µg/ml.Hemolytic activity tests revealed only a hemolytic effect of only 13% at the highest concentration of 50 µg/ml. As a result of the thesis work, it has been found that all of the skin secretion of L.vulgaris has bioactivity in different amounts. At the same time, this skin secretion has given rise to results that can be taken as part of drug development work to be carried out in terms of content.
