Summary
Idiopathic pulmonary fibrosis (IPF) is a aging-related, chronic lung disease whose pathogenesis is unclear, has no effective treatment. One of the triggering factors in cell aging is oxidative stress and it is known to have a role in IPF. In this thesis; the protective effect of 3,4-lacto-cycloastragenol molecule in terms of its antioxidant properties was evaluated in mice lung fibrosis model. Fibrosis was examined by Hematoxylin-Eosin and Masson Trichrome staining, immunohistochemical staining for TGF-ß1, Type I collagen, Ki-67, ?-H2AX markers. Besides, activity analysis of catalase and SOD enzymes, measurement of total oxidant, total GSH and MDA levels and western blot analysis for Nrf2 were performed. Last, in histology analysis, it was determined that three different doses of the molecule provided a prophylactic effect against the progression of fibrosis compared to the BLM control group. However, it was observed that only the molecule applied in the high dose increased Nrf2 nuclear translocation and decreased TOS.
