PREPARATION, OPTIMIZATION AND APPLICATIONS OF NANOFIBERS FOR OPTICAL-BASED SENSOR PLATFORMS

Project no. 22714 · Status: Kapanmış · 2021

Summary

Intense studies have been conducted on immobilizing biomolecules without reducing their biological activities while maintaining proper orientation. It is crucial to select the appropriate material for immobilization carefully. To improve the sensitivity and selectivity of the prepared analysis systems, it is necessary to increase the interaction interface between the biomolecule-bound surface and the analyzed sample matrix, that is, to improve the interactions in the microenvironment by using nanostructured materials. In particular, nanofibers prepared using hydrophilic and hydrophobic polymers provide advantages in immobilising biological molecules.

In this project, four different sensor systems were developed: enzyme, immunosensor and aptasensor. Nanofibers based on hydrophobic/hydrophilic polymers were produced by electrospinning. First, the preparation conditions of nanofibers were optimized. The aim was to determine the best surface for interaction with biological molecules. Optimization of nanofibers: It covers morphological and structural characterization and, finally, biomolecule immobilization and its applications in the optical determination of target molecules, which are biomarkers, especially in evaluating some diseases. Recognition surfaces were prepared by immobilizing biomolecules on the surface of the prepared nanofibers. Quantitative determinations were made by adding target analytes and their relevant bioconjugates to the developed measurement systems. New analysis systems have been developed that have the potential for non-invasive application and are suitable for use in point-of-care analyses. Artificial sweat, artificial serum and artificial saliva were used to test their suitability for analysis in real samples. Afterwards, in vitro toxicity tests were performed to test their suitability for use as a tattoo. The results obtained have become a tool for new projects. This project will support and enrich research for point-of-care analyses. In addition, it is thought that this project will positively strengthen interdisciplinary relations and support future multidisciplinary projects.
