The Interaction Of Pemetrexed Loaded Nanoparticles And Propolis In Cancer Treatment
Project no. 23666 · Status: Kapanmış (Yayın bekleniyor) · 2022
Summary

The longstanding fight against lung cancer places a huge burden on public health and health economics. Many efforts are therefore being made to prevent and manage lung cancer. The most widely studied area is nanotechnology, where cancer cells are targeted through different strategies using nanocarriers. However, the use of natural substances with therapeutic potential is also a frequently preferred method. This study aimed to understand the effect of propolis, a popular natural product in apitherapy, on cells when used in combination with pemetrexed, a chemotherapy drug. The characterization of the synthesized nanoparticles (MSNs) was carried out by differential analysis. The average size distribution of MSNs determined by DLS analysis was 140.1 nm and the particles were found to be monodisperse. The size obtained by Scanning Electron Microscopy (SEM) was in the range of 68-75 nm and the particles were spherical. Pemetrexed was loaded into mesoporous silica nanoparticles to reach the target in a more protected manner. The loading capacity efficiency was calculated as 79% and the release of the drug started before the 24th hour.

In order to understand the effects of the combined treatment, cytotoxicity and apoptosis studies were performed in vitro in BEAS-2B and A549 cell lines. IC50 values of the sample groups were calculated as a result of the cytotoxicity study. Propolis was found to have the lowest IC50 value (4.7 µg/mL) and the highest cytotoxic effect in A549 cell line. The lower IC50 value of propolis in healthy/normal cell lines compared to cancer cell lines is an important sign of its selectivity and therapeutic potential in cancer cells. The cytotoxic effect of the MSN-PEM-propolis sample group was also found to be higher, but should be supported by further experiments and clinical studies.
