Investigation of the Biological Properties of Different Pulp-Capping Materials

Project no. 20905 · Status: Kapanmış · 2019

Summary

Aim: To evaluate the biocompatibility, inflammatory response and antimicrobial activity of different pulp-capping materials.

Materials and Methods: Five pulp-capping materials (ProRoot MTA/Dentsply, Biodentine/Septodont, Dycal/Dentsply, TheraCal LC/Bisco, calcium hydroxide/Saver-Pyrax) and an experimental-material (calcium hydroxide+dentin chip) were examined. The effects on cell viability were investigated via WST-1 in L929 cells. The inflammatory response was determined by the change in monocyte chemoattractant protein 1 (MCP-1/CCL2) and macrophage inflammatory protein-1α (MIP-1α/CCL3) levels. The antimicrobial activity against oral-microorganisms (Streptococcus mutans, Lactobacillus acidophilus and Enterococcus faecalis) was examined by agar-diffusion.

Results: The lowest cell viability at 24 and 48 hours was detected in ProRoot MTA and Dycal (p<0.05). The cell viability of the calcium hydroxide+dentin chip was similar with control and Biodentine at 24 hours. The cell viability of calcium hydroxide+dentin chip group decreased at 48 hours, but it was higher than ProRoot MTA and Dycal (p<0.05). The highest MCP-1 value was detected in the ProRoot MTA and TheraCal LC at 24 and 48 hours compared to the control (p<0.05). The MIP-1α values of the experimental groups were not different from the control in both time intervals. The MIP-1α level of calcium hydroxide+dentin chip was lower than ProRoot MTA and Biodentine in 48 hours (p<0.05). In the agar diffusion, a limited inhibition zone (16±1 mm) against Streptococcus mutans was detected only in the Dycal.

Conclusion: The materials exhibit different properties according to their content. The calcium hydroxide+dentin chip has advantages to be considered as a pulp-capping material, and further researches are necessary to support the data.
