Evaluation of Anti-Inflammatory Effects of Different Endodontic Medicaments on Human Macrophage Cells

Project no. 349 · Status: Kapanmış · 2018

Summary

Aim To assess the effects of calcium hydroxide (CH), chlorhexidine (CHX), cetylpyridinium chloride (CPC) and benzydamine hydrochloride (BNZ) medicaments on IL-1ß, IL-6 and TNF-? expression levels of macrophage cells. Methodology Gel formulations were prepared using CH, CHX, CPC, BNZ and Chitosan medicaments. THP-1 cells (human monocyte cells) were differentiated into macrophages by 24-hour incubation with phorbol 12-myristate 13-acetate (PMA) and then, lipopolysaccharide-activated cells were used to test the anti-inflammatory potential. IL-1ß, IL-6 and TNF-? levels in supernatants were determined using enzyme linked immünosorbent assay (ELISA) kits. The data were examined with One-way ANOVA, Dunnett’s Multiple Comparison Test and Student’s t test (P=0.05). Results The cytotoxicity assay revealed that CH and CH+Chitosan, Chitosan did not affect the cell viability. The highest cell viability was observed with BNZ, BNZ+Chitosan, CHX+Chitosan, and CHX, respectively. Whereas CPC, doxorubicin and CPC+Chitosan have significantly decreased the cell viability. The lowest IL-1ß values were observed with BNZ+Chitosan 50 µg/ml (P<0.05), whereas all CPC combinations significantly higher IL-1ß values than the level of IL-1ß observed with inflammation (P<0.05). The lowest IL-6 values were observed with BNZ+Chitosan50 µg/ml. Chitosan 10 µg/ml, CH 50 µg/ml and CH+Chitosan 50 µg/ml had significantly higher values than the level of IL-6 observed with inflammation. All test samples had significantly lower TNF-? values than the level of TNF-? observed with the inflammation. The lowest TNF-? values were observed with BNZ 50µg/ml and with BNZ+Chitosan 50µg/ml. Conclusions BNZ+Chitosan combination has revealed promising anti-inflammatory effects, nevertheless the present findings need to be examined in clinical endodontic conditions.
