Summary

Recently, phenolic compounds as natural sources have been (Quercetin, Kaempferol, Ellagic acid and Myricetin) suggested to use in treatment and chemoprevention of prostate cancer. Since rosehip includes these molecules in high concentration, we aimed to investigate the possible anti-proliferative and apoptotic effect of rosehip tea on the prostate cancer cell line. We determined the flavonol content of rosehip tea which was prepared in different temperature and time interval, then the antiproliferative effect of tea samples were determined through by addition of tea samples into prostate cancer cell line (VCaP and LNCaP). The phenolic compound content of the teas was analyzed using the LC-MS / MS system. For the cell culture study, 7 minutes brewed fruit, 7 minutes brewed commercial fruit, 5 minutes brewed and shredded fruit, 3 minutes brewed commercial fruit extracts were selected with the highest quercetin, ellagic acid, myricetin and kaempferol levels. The tea samples were prepared in relevant concentration to their IC50 values and then added into VCaP and LNCaP cell line. Quercetin was more effective in LNCaP cell than VCaP cell. The antiproliferative effect of rosehip teas on VCaP cells was slightly stronger than LNCaP cells. The tea extracts obtained from flavonels, quercetin, Myricetin, Kaempferol, Ellagic acid which have the highest amount of different phenolic substances in the obtained teas have two different VCaP and LNCaP apoptotic effects on prostate cancer cell line by using ELISA kit method by assessing Caspase 3, Caspase 8, Caspase 9, TRAIL, APAF-1 levels. AnnexinV-FITC levels were determined by Flow cytometery method. Our data clearly indicated that rosehip tea as a natural source of flavonols might be proposed in treatment of prostate cancer. However, the extracted amount of phenolic molecules is attributed with preparation techniques, so it is suggested to determine the optimum conditions and achieve highest concentration of flavonols.
