PHYLOGENETİC DİVERSİTY ANALYSİS OF TRİCHODERMA SPECİES USED AS BİOCONTROL AGENTS
Project no. 700 · Status: Kapanmış · 2015
Summary
In this study, it was aimed to carry out molecular identification of 11 isolates of Trichoderma spp. which known to be a biological control agent and provided from the Culture Collection of Fungi of the Bioengineering Department of Ege University. For this purpose, the Internal Transcribed Spacer ( ITS) and Elongation Factor (EF) gene region of Trichoderma spp. was amplified with Polymerase Chain Reaction and then amplicons was sequenced. By comparing the sequences of the ITS and EF region to the sequences deposited in NCBI-GenBank using BLAST program, all of the isolates were identified to species level as T. citrinoviride (n=6) and T. atroviride (n=5). In addition, TrichOKEY and TrichoBLAST search tools was used to assess the reliability of NCBI- BLAST. Then, all nucletiode sequences belong to the ITS and EF genes have been deposited at NCBI-GenBank and the phylogenetic trees were constructed with MEGA v.6. using nucletiode sequences. This study describes polymorphic region in the nucleotide sequence of the ITS and EF genes for T. citrinoviride (n=6) and T. atroviride (n=5). Therefore, the unique primers will be designed for T. citrinoviride and T. atroviride. As a result, it will be develop innovative approaches for the fast and low cost molecular identification of the Trichoderma spp.
