Summary

Ovarian cancer is a lethal gynecological cancer and is the most common cause of cancer death among women, as it usually occurs in advanced stages. Depending on the degree and progression of the disease, surgical treatment, radiotherapy, or chemotherapy treatments are applied. Cisplatin is one of the chemotherapeutic agents commonly used in chemotherapy treatment, but chemotherapy resistance may develop after long Cisplatin treatment.

NF-κB is one of the important transcription factors that govern many processes in the cell. As a major cell survival signal, NF-κB plays a role in multiple steps in cancer cell resistance to chemotherapy and radiotherapy. It is hypothesized that enhanced drug resistance can be reversed by inhibiting NF-kB activation in cancer cells with various anticancer agents.

Resveratrol, a natural polyphenol compound, is reported to have anticancer activity in various cancer cells. It is abundant in a variety of foods, including grapes, strawberries, and peanuts. Many studies have shown that resveratrol has anti-tumor, anti-inflammatory, and anti-atherosclerotic effects through various pathways in the cell. In addition, it has been reported to stop the cell cycle, induce apoptosis and autophagy, and inhibit cell proliferation. In addition, studies are showing that it increases the sensitivity of drug-resistant cancer cells to drugs by inhibiting the NF-kB signaling pathway responsible for the expression of many genes in the cell. However, since there is no literature indicating whether Resveratrol sensitizes cells to Cisplatin by inhibiting cell cycles and NF-kB activity in the A2780 cell line with cisplatin resistance, we think that our research will have a unique value.

In this study, we comparatively examined the effects of Resveratrol on intracellular molecular mechanisms such as cell cycle, cell proliferation, and apoptosis, depending on the activation changes on the NF-κB signaling pathway, using the ovarian cancer cell line A2780 with and without Cisplatin resistance. According to our findings, it was found that Resveratrol inhibited proliferation and showed a cytotoxic effect in A2780 and A2780cis cell lines. According to the cell cycle analysis, apoptosis analysis, and RT-PCR analysis we performed afterward, we found that Resveratrol had no effect in line with the literature. Since the findings we obtained did not reveal a significant result, we think that in our future studies, it may be useful to apply Resveratrol to cells in combined doses with Cisplatin and to measure protein assays and phosphorylation levels to determine its effect on NF-kB expression.
