Isolation Purification and Structural Determination Studies on Propolis Samples Collected from Different Regions of Turkiye

Project no. 23101 · Status: Kapanmış · 2021

Summary

Propolis, a natural mixture obtained by bees mixing resins collected from plants with their saliva, stands out with various biological activities and its importance in industry is increasing day by day. However, the biggest problem in propolis usage is standardization. Standardization issues of propolis vary according to geographical and plant sources. This diversity is directly related to the chemical content of propolis and determines the product quality. Therefore, it is crucial to accurately define the chemical content of propolis. Spectroscopic chemical content screening studies can be conducted for this determination. However, such screenings generally remain limited to the compounds known in the literature. Yet, new pharmacologically active compounds are continuously discovered in studies on natural products. Propolis isolation studies have been quite limited worldwide to date, with only one in our country. The deficiency in identifying the rich contents of propolis in our country needs to be addressed.

Therefore, our study focuses on the isolation, purification, and structural determination of propolis collected from Rize (Çamlıtepe and Kaçkar) and Gümüşhane (Kelkit) districts for the first time. Damaran and flavonoid-rich propolis were studied separately. The structures of the isolated compounds were determined using FTIR, 1H NMR, 13C NMR, COSY, HSQC, and HMBC methods. A new compound, damaran-3-one-30-hydroxy-12-acetyloxy-24-methylene, was obtained from Çamlıtepe and Kaçkar propolis. Additionally, four compounds were obtained from Kelkit propolis, one of which is pinosembrin. The other three compounds will undergo NMR analysis again due to insufficient quantity to obtain measurements at higher frequencies. It is believed that this study is important for revealing the structure-biological activity relationship in future stages.

Keyword: propolis, flavonone, flavon, dammarane
