INVESTIGATION OF THE EFFECTS OF NO-MEDIATED REACTIVE NITROGEN SPECIES (RNS) ON THE CELL WALL MODULATION
Project no. 436 · Status: Kapanmış · 2017
Summary
The seeds of creal plants are generally planted under nearly a few centimeters of the soil surface. However, in order to be grown, the seeds are planted deeper in the higher layers of soil that has not enough moist. Thus, plants can take the sufficient water for germination and seedling through moistened soil by the underground water. It is necessary that the plant should adapted to deep sowing tolerance. The deep sowing tolerance of wheat occurs depending on the first internode elongation. The key part of elongation mechanism is loosing of the cell and accordingly elongation. Although there are some studies done related to the effects of hormons as gibberellin and reactive oxygen species as hydroxyl radical on this mechanism, there is no study done about the impact of RNS molecules on the first internode elongation. In addition, recently, the importance of the relation between ROS and enzymes depending on this in the physiological process has been carried out. In the master thesis presented within this context, c-PTIO (NO and NO synthase inhibitor) is applied to Tir wheats that have deep sowing tolerance and the plants are grown both normal and deep sowing conditions. The first internode, coleoptile and growth length of 10 days old plants are measured and both the answer to deep sowing and NO sensibility are defined by harvesting. Besides, by observing some antioxidative enzymes activities in the first internodes (POX, CAT) and by defining the enzyme activities of CWPOX and NOX that are producing ROS. It is aimed to clarified that the effects of RNS on the first internode elongation and the relation of ROS molecules in this elongation.
