Summary
This study was conducted to determine the prevalence and molecular characterization of Cucumber mosaic virus and Tomato spotted wilt virus that cause infections in tomato and pepper plants growing in İzmir province including Ödemiş, Bayındır, Torbalı, Menderes and Tire districts. 18 samples from Ödemiş, 20 samples from Bayındır, 117 samples from Torbalı, 9 samples from Menderes and 22 samples from Tire were collected from tomato plants having virus-like symptoms. 10 samples from Ödemiş, 13 samples from Bayındır, 11 samples from Torbalı, 43 samples from Menderes and 29 samples from Tire were collected from pepper plants. Out of collected 186 tomato samples 40 (21.50%) described as CMV and 40 (21.50%) as TSWV as a result of DAS-ELISA test. Out of collected 106 pepper samples 27 (25.47%) detected as CMV and 68 (64.15%) as TSWV. Some regions on RNA 3 of CMV and S RNA of TSWV of DAS-ELISA positive samples were amplifed by RT-PCR with specific primers. Similarity rates of tomato CMV isolate varied between 98-95% identity with other isolates from genebank and pepper CMV isolate varied between 100-98%. Pepper TSWV isolate showed identity varied between 99-41% with other isolates from genebank.
