Preparation of Amnion Membrane Extract and Analysis of Its Biochemical Content
Project no. 21402 · Status: Kapanmış · 2020
Summary

Human amniotic membrane (iAM) is the inner layer of the placenta consisting of epithelium, basement membrane, and connective tissue. It has an extracellular matrix rich in cytokines, growth factors, collagen, and laminin. These characteristics of iAM can provide recovery with the proliferation of epithelial cells, suppressing inflammation, scarring, and angiogenesis. It is widely used in resistant pathologies such as persistent epithelial defect (incidence 30%), pterygium (prevalence: 15%), and limbal stem cell deficiency (prevalence: 1/10000) have high prevalence and incidence in many fields of medicine and ophthalmology. In ophthalmology, iAM is often prepared by cryopreservation. It is then used as “iAM transplantation”. After transplantation, it is resorbed after it remains on average 2-8 weeks in the eye,. This form of use requires surgery and cannot be used as long as desired. It has limitations such as the inability to perform objective clinical observation due to its semi-opaque nature. preparation of amniotic membrane extract (AME) in order to reduce these limitations. The idea of applying this in clinical practice has arisen. Studies on the production of AME and its usage areas are continuing actively all over the world, and for the final product, patent applications and patent records are available. Although obtaining AME is an important research area, there is no standardization of preparation, storage and sterilization methods. The aim of this study is to prepare AME for the first time in Turkey, to determine the storage and sterilization conditions, to establish a national standard on issues, and to determine the amount of growth factors in it. The development of AME eye drops in this way will provide a basis for future experimental and clinical studies. For this purpose, 10 amniotic membranes are going to be homogenized and biochemically analyzed and then AME is going to be lyophilized and sterilized.
