Türkiyede Rotavirüs ve Norovirüs Gastroenteritine Yatkınlıkta FUT2 Gen Polimorfizmlerinin Araştırılması
Project no. 20725 · Status: Kapanmış (Yayın bekleniyor) · 2019
Summary
Introduction: Norovirus and rotavirus are the leading causes of diarrhea worldwide. Both viruses recognize and bind to histo-blood group antigens (HBGA), which are oligosaccharides found on epithelial cells of the gastrointestinal and respiratory tracts. Expression of HBGA in the gut is controlled by the fucosyltransferase 2 (FUT2) gene. A number of of polymorphisms in the FUT2 gene have been shown to reduce the expression of HBGA, and these so-called 'non-secretory' alleles have been shown to be protective against rotavirus and norovirus infection. In this study, it is aimed to investigate FUT2 gene polymorphisms in susceptibility to rotavirus and norovirus gastroenteritis in Turkey. Material-Method: This study included 110 children (between 0-10 years old) diagnosed to have rotavirus or norovirus gastroenteritis by PCR test and 107 healthy children (between 8-10 years old) with no history of rotavirus or norovirus gastroenteritis. . Demographic and clinical data of both groups were recorded. Sequence analysis of the FUT2 gene was performed on a next generation sequencing platform. All SNPs (single nükleotid polymorohisms) found in the FUT2 gene were evaluated in the patients and control group. Results: In our study group, 57 (52%) were male and 53 (48%) were female. Rotavirus and norovirus were detected in 93% and 7% of patients. Fifty of 110 patients were hospitalized.. 58% of them were mildly dehydrated, 36% moderately, and 6% severely Among the 14 SNPs evaluated, the presence of T allele for the rs281377 (390C>T) increased the risk of disease 1.58 times (OR=1.58 [1.08-2.32]). Fort the c.*232A>G polymorphism , the presence of G allele increased the risk of the disease 3.94 times (OR=3.94 [1.10-14.18]). The rs601338 (461G>A) polymorphism was protective in the homozygous condition (OR=0.36 [0.16-0.78]). Conclusion: In this study, the entire FUT2 gene was sequenced for the first time in Turkish patients with rotavirus and norovirus gastroenteritis, and it was determined that the G allele in a SNP (c.*232A>G) in the 3'UTR region of the gene increased the risk of disease 3.9 times. It was found that rs601338 polymorphism, which is known to be associated with nonsecretory phenotype in homozygous condition in previous studies, also protects from the disease in our population. The frequency of FUT2 gene polymorphisms differs between populations. There is no study has been conducted on this issue in Turkey reveals the importance of the study.
