MUTAGENIC INVESTIGATION OF IZMIR INTERNAL GULF SEDIMENT WITH AMES GENOTOXICITY TEST

Project no. 20553 · Status: Kapanmış (Yayın bekleniyor) · 2019

Summary

With its aquatic organisms, Izmir Bay is one of the most important ecosystems of Aegean Region. With its harbor, it is among the cities of commercial and touristic interest. Importance of detecting chemicals with impacts on human health and environmental impacts is well known. Impacts of environmental pollution in the aquatic environments, especially impacts of pollution with mutagenic and carcinogenic substances on human health is an important research object. Thus, it is required incorporate short-term biological research methods with molecular chemical analysis methods. Ames’s assay, which is a practical and validated method to detect toxic substances in the environmental samples, is one of the most important genotoxicity assays continuing to be relevant. By means of Ames’s assay, it is possible to determine mutagenic potential of several chemicals, environmental pollutants, sediments, and waste waters. For this reason, Ames’s mutagenicity assay without S9 enzyme using TA98 and TA100 strains of Salmonella typhimurium was done at 4 different concentrations (125 µg, 250 µg, 375 µg, and 500 µg) in order to detect direct mutagens in the sediment samples from 6 stations on Inner Bay of Izmir. Based on the results of Ames’s assay, it was found that especially Turan Area (Station #3, on which shipyard is located) among 6 stations on Izmir Bay was under mutagenic and toxic effect and Bostanli Area (sta.6) was under intense toxic effect. Turan Area was under effect of base-pair and frameshift mutagens.
