OPTIMIZATION OF MORPHOLOGICAL AND MOLECULAR DIAGNOSTIC PROTOCOLS FOR ASPERGILLUS AND PENICILLIUM SPECIES ISOLATED FROM ORGANIC RAISINS
Project no. 21671 · Status: Kapanmış (Yayın bekleniyor) · 2020
Summary

The Aegean Region is one of the leading geographical areas of Turkey in raisin production. However, raisins are very susceptible to contamination by some mycotoxin-producing fungal organisms during the production and drying process. In a previous study, mold fungi consisting of Aspergillus section Nigri member and Penicillium species were isolated from grape samples grown, processed and dried in accordance with organic farming principles. It is not possible to diagnose these molds based only on morphological diagnostic criteria. In this study, both morphological and molecular methods were tried to be used together in order to make accurate, rapid and reliable identification of the fungi causing the mold load detected in organic raisins.

DNA barcoding of 35 of the 40 fungal isolates constituting our study material was performed using Bt2a-Bt2b and CMD5-CMD6 primers that amplify Btubilin Cadmodulin genes. The obtained bidirectional sequences were made contig and 9 different species were identified using the NCBI database. Among these, the most frequently obtained species was Aspergillus uvarum with 12 isolates. This was followed by A. tubigensis with 9 isolates. The other species were Penicillium chrysogenum, A. fumigatus, A. terreus, Talaromyces siamensis, A. acculeatus, A. fructus, A. welwitschiae , respectively.
