Investigation of deaminoneuraminic acid antigen and monoclonal antibodies in blood and urine samples of cancer patients

Summary

The number of studies on the determination of new molecules to be used in the diagnosis and treatment of cancer is increasing day by day. Previous studies of our research group focused on deaminoneuraminic acid, a molecule that can be used in cancer diagnosis. This molecule is 2-keto-3-deoxy-D-glycero-D-galactonononic acid (Deaminoneuraminic acid); It is an important member of the sialic acid family, formed by the addition of a hydroxyl group instead of the aceto-amide group at the C5 position in a common Sia (N-acetylneuraminic acid-Neu5Ac). According to the previous results, the molecule is found at a higher rate in prostate and breast cancer cell lines than in normal epithelial cell lines. These findings suggested that the molecule known to be found on the tumor surface can be used for early diagnosis in cancer patients. Here, it was aimed to investigate the presence of deaminoneuraminic acid antigen in blood and urine samples taken from cancer patients. Using HPLC methods together with biostatistics (chemometry, principal component analysis, PLS-DA), 2-keto-3-deoxy-D-glycero-D-galacto-nononic acid antigen levels were quantitatively determined in the blood and urine of randomly selected cancer patients. KDN antigen levels showed significant differences in blood and urine samples from cancer patients and healthy volunteers. In the light of the results of this project, further characterization of the KDN monoclonal antibody and elucidation of the structure of this antibody are required in future studies. Thus, KDN antigen/antibody can be used as a biomarker for early detection of cancer and rapid kits can be designed.
