Determination of transcriptome change caused by green mold disease agent Trichoderma spp. in edible mushroom
Project no. 20953 · Status: Kapanmış (Yayın bekleniyor) · 2020
Summary

Mushroom production is highly affected by green mold disease, which causes serious losses worldwide. Molecular characterization of 56 Trichoderma isolates obtained from mushroom production areas in Aegean and Marmara regions was performed. A phylogenetic tree was constructed based on ITS and tef1 sequences. T. aggressivum, T. europaeum, T. pleuroticola, T. virens, T. harzianum and T. afroharzianum isolates were identified. The tef1 marker was found to be better at detecting genetic variations to the ITS marker. Agaricus bisporus-Trichoderma spp. growth inhibition was seen between 50% and 75% in the dual test. An in vivo study was performed to determine the effects of two Trichoderma isolates (3/9 and 7/5) with the highest lytic and mycoparasitic activity on yield and mushroom quality parameters. As a result, T. aggressivum f. agressivum and T. agresivum f. europaeum isolates significantly inhibited the growth of A. bisporus in vitro and in vivo (p < 0.05). RNA-seq analysis was performed from control pin, control cap and cap infected with Trichoderma species. As a result of RNA-seq analysis, it was determined that the gene expression of genes named 114133,121394,151828 increased in the control cap compared to the control pin. On the other hand, it was determined that the gene expression of the genes named 176453,194666 decreased in the control cap compared to the control pin. It was determined that the gene expression of 205013 and 193671 genes increased in the infected cap compared to the control pin. The corresponding metabolism, functions and pathways for these genes were determined in the KEGG database. Validation of RNA-seq analysis with real time has been achieved.
