Investigating of Circulating miRNAs (Let-7, miR-71, miR-10) According to Cyst Stages in Patients with Cystic Echinococcosis

Project no. 20565 · Status: Kapanmış (Yayın bekleniyor) · 2019

Summary

Cystic echinococcosis (KE) is a parasitic disease which larval form of Echinococcus granulosus causes cysts in various organs, especially in the liver. The World Health Organization (WHO) has classified cyst types from CE1 to CE5. This classification is important for the activity of the cyst and follow-up of the patient. MicroRNAs (miRNAs) are 18-22 nucleotide-length RNAs that regulate their expression by inhibiting translation or manipulating transcription. Studies have shown that miRNAs are stable in serum and plasma. In our study, it was aimed to investigate 3 of miRNAs (egr-let-7, egr-miR-71 ve egr-mir-10), which were previously discovered according to literature research in E.granulosus, in blood samples of patients with CE and to compare them according to cyst types. Blood samples of CE patients (n: 50) who had cyst classification on ultrasonographic examination and healthy volunteers (n: 20) were evaluated with ELISA and Western Blot (WB). Then, for the miRNA analysis, the evaluation of the target 3 miRNA was performed using the Quantitative Real Time PCR (qRT-PCR) method. miRNA expression levels were calculated according to the cyst classification by calculating according to the 2 -??CT method. Thirty-one (62%) of the patients were women and 19 (38%) were men and their ages ranged between 8-73 years. According to the ELISA results, 41 (82%) samples were positive, two (4%) samples were found at threshold and 7 (14%) samples were found negative. CE-specific bands were detected in the samples of patient group with WB method. Healthy control group samples were found negative in terms of CE by ELISA and WB methods. In patient samples, amplification was detected in 27 (54%), 5 (10%) and 13 (26%) for egr-let-7, egr-miR-10 and egr-miR-71, respectively. No amplification was detected in the healthy control group. The expression level of the patient group was significantly different in miRNA expression levels compared to healthy controls (p <0.05). According to cyst classifications, statistically significant difference was found in egr-miR-71 in CE1 and egr-let-7 in CE2 (p <0.05). As a result; in our study, it was shown that miRNAs from E.granulosus can be detected in plasma samples of CE patients. According to the cyst classifications, it is promising to use egr-miR-71 in CE1 and egr-let-7 in CE2 for early diagnosis and follow-up of CE patients. Our study is the first study in Turkey for the detection of miRNAs in blood samples of CE patients so far.
