Determınation of the Molecular Characterization of Diyarbakır Local Eggplant Genotypes
Project no. 21770 · Status: Kapanmış · 2020
Summary

In this study, it is aimed to determine the molecular characterization of eggplant genotypes. 37 local genotypes collected from districts and villages of Diyarbakır province; 1 local genotype from gene bank; 4 commercial variaties used in provience; 14 genotypes from commercial seed company and 9 genotype obtained from Asian Vegetable Research and Development Center were used as plant material (total 65 genotypes). Seeds of genotypes (15 seeds per genotype) were sown manually in peat medium in EPS seedling tray with 128 cells. The trays were kept in the germination room at 22-24ºC day/night and 85% relative humidity for 3 days in dark conditions. After the seeds germinate, the trays were taken to the seedling production greenhouse and kept there until the seedlings reached planting size. When the seedlings had 3-4 leaves, leaf samples were taken for DNA extractions. Leaf samples were dissected with the help of Tissue Lyser. DNAs of genotypes were obtained by applying the protocol included in the plant DNA extraction kit (DNeasy Plant Mini Kit-Quigen) to the fragmented samples. For this purpose, SSR markers were used and the markers were selected from the relevant literature and DNA marker database for vegetables belonging to NARO-Institute of Vegetable and Ornamental Sciences. Evaluations were made according to PIC and allele number and 28 SSR markers (primer) were selected to be used in determining genetic diversity. Scores of genotypes based on SSR primers; It was analyzed using the NTSYS-PC package program. After the similarity coefficients of the genotypes were created using the Jakkard method, their dendograms were created with the arithmetic mean and the UPGMA method. After the correlation matrix was created with the SIMINT module of the program, PCA analysis was performed. As a result of the comparisons made on the bands obtained from the scored primers, a total of 72 polymorphic markers were obtained from 28 primers, and it was determined that all of the primers used were polymorphic. It was observed that the similarity rate of the genotypes varied between 29% and 100%, and all genotypes were grouped under 6 groups and most of the genotypes were in group A. As a result of the research, it was found that the eggplant genotypes collected from the region showed genetic variation, but there were similar groups; It has been determined that the current variation is important for genetic diversity and that the obtained data can be used in breeding studies.
