ACTIVITY-GUIDED PURIFICATION AND DETERMINATION OF BIOACTIVE PEPTIDES FOR THE TREATMENT OF TYPE 2 DIABETES FROM BARLEY (Hordeum vulgare L)
Project no. 1097 · Status: Kapanmış · 2017
Summary
Diabetes mellitus is multifactorial and chronic disease which is characterized with hyperglycemia and insulin resistance. Because of increasing numbers of people exposed to the disease in the world population, preventing the formation of the disease and developing new treatments has become a necessity. In recent years, there is an increased interest in the study of compounds from many natural sources that can be used in diabetes treatment. The therapeutically active compounds obtained from natural sources are advantageous in terms of low toxicity, high stability in physiological conditions instead of those of synthetic origin and they have various chemical structures for the design of new drugs. For this purpose, many natural sources have been investigated for their biological activity and there are a number of studies that focus on endogenous and endogenous peptides. Many studies have reported that peptides from various food sources possess bioactivities, including antihypertensive, antioxidant, anticancer, antimicrobial, and opioid activities as well as immunomodulatory and cholesterol-lowering effects. Bioactive peptides are inactive within the sequences of the parent proteins. They are released by enzymatic hydrolysis and then they may exert various physiological functions. In this context, germinating barley is selected as endogenous peptide source in this project. The aim of this work was to purify and identify peptides with biological activities (DPP4, ?-glucosidase inhibition, metal chelating/antioxidant properties) from germinating barley. Results showed that germinating barley represents a good source of bioactive peptides. Purified peptides were evaluated by means of their biological activity effectness IC50/EC50 and peptide sequences. The results indicate that 2 peptides are novel and high effectiveness for deglycation activity and, can be the source of potential drug effects in the prevention of diabetes and its complications. It is clear that it is necessary to carry out in vitro and in vivo kinetic studies of these peptide preparates. Keywords: Type 2 Diabetes mellitus, bioactive peptides, Barley (Hordeum vulgare L)
