Summary

ASSESMENT OF THE EFFECTS OF GREEN SYNTHESIZED COPPER OXIDE NANOPARTICLES (CuO NP) ON SEED VIGOUR AND SEEDLING DEVELOPMENT OF TOMATO HASEKİ, Selin MSc. in Department of Biology Supervisor: Prof. Dr. Lale AKTAŞ December 2021, 60 pages CuO nanoparticles (CuO NP) have the potential to be used in many agricultural applications as pesticide, herbicide, fertilizer and/or plant growth regulators due to their easy synthesis and significantly increased efficiency at nanoscale. The aim of this study is green synthesis and characterization of copper oxide nanoparticle and to determine its effects on germination and seedling growth on tomato (Lycopersicon esculentum). CuO NPs were prepared by using laurel (Laurus nobilis L.) leaf extract and the nanoparticle formation was confirmed by UV/VIS spectrophotometric measurement with characteristic peak values between 210 -340 nm. The FT-IR analysis results showed that the nanoparticles were coated with phytocontent, and the Zeta potential results showed that the nanoparticle was stable with a value of -22 mV. SEM-EDS imaging revealed that CuO NPs were spherical in shape with an average size of 83 nm, and the element percentage of the particle was Cu (45.04%). According to the results of ICP MS analysis, the total copper concentration in the CuO NP solution was determined as 583 ppm (mg/kg). The priming process of tomato plants was carried out by applying green synthesized CuO nanoparticles at 5 different concentrations (6.25, 12.5, 25, 50 and 100 mg L-1) to the seeds for 48 hours. After the germination trials, nanoprimed seeds were grown under greenhouse conditions for 21 days. Results showed that priming with 25 mg L-1 CuO NPs increased germination percentage by 17.5% and seedling vigour by 26.79% compared to control. However, high concentrations of CuO NP application to seeds suppressed seedling growth; It was determined that 100 mg L-1 CuO NP inhibited root and stem elongation by 33.18% and 35.6%, respectively, and it caused 37.5% and 45% inhibition in fresh and dry weights, respectively. The results revealed that the green synthesized CuO NPs could penetrate into the seed by nanopriming process and their effects on seed germination and seedling growth could vary depending on the amount of application.
