Investigation of the Interaction of Bioactive Natural Compound Ruscogenin with Model Membranes at the Molecular Level by Biophysical Methods
Project no. 22592 · Status: Kapanmış · 2021
Summary
Biological membranes are one of the most important cell structures surrounding cells and cell organelles. It plays an important role in almost all cellular events, as it forms a barrier between the inside and outside of the cell. Because biological membranes contain so many components, it is difficult to understand the interactions between components and additives. Therefore, studying with model membranes in which we can change the number and concentration of their components becomes important in explaining the events in biological membranes. Generally, these systems are spherical vesicles in which lipids form a bilayer, consisting of mixtures containing one or more phospholipids of natural or synthetic origin. Model membranes have been the subject of extensive research as a model for biological membranes. In this study, the physical interaction of ruscogenin, a bioactive antioxidant in steroid structure, with model membranes consisting of phospholipids, that is, its effect on membrane phase transition, membrane arrangement and membrane dynamics, as well as the thermal changes that occur with the addition of ruscogenin to model membranes, depending on temperature and concentration, were investigated using FTIR and DSC techniques. The results from this study have helped us to have information about the physiological effects of bioactive compounds such as phospholipids and ruscogenin on humans. The findings have contributed to the elucidation and interpretation of the molecular mechanisms of the biological effects of ruscogenin on membranes.
