MOLECULAR EFFECTS OF NOCODAZOLE AND ASTAXANTHINE IN BREAST CANCER CELL LINES, OSIMERTINIB AND FUCOIDANE IN OVER CANCER CELL LINES
Project no. 79 · Status: Kapanmış · 2017
Summary
Breast and ovarian cancers are among the main cancers seen in women. Drug treatment is among the important options for both, as in other cancers. However, new conncepts are on the agenda in drug treatment. For this reason, the issue of the combined use of alternative agents in the treatment process is frequently discussed and alternative drugs are frequently used in studies. In our study, for MCF-7 and MDA-MB-231 breast cancer cells, astaxanthin, which is our natural therapy option, was combined with our chemotherapeutic agent nocodazole; similarly, it was investigated whether fucoidan, which is our supportive therapy option for OV-CAR and SK-OV ovarian cancer cell lines, combined with osimertinib creates a synergistic effect in terms of cytotoxicity and apoptosis. While the cytotoxic and apoptotic effects of both agents and their combinations on breast cancer cells were evident, the effects of fucoidan on ovarian cancer cells were evident, and the effects of osimertinib and its combinations were not found to be significant. In addition to these, in our study, changes in genes, related to signal transduction, cell cycle and pathways that are important in cancers for expression changes that occur after drug administration are presented. Here, in parallel with cytotoxicity and apoptosis experiments, there was a decrease in the expression of genes in favor of cancer in both agent and combination groups in breast cancer cell lines, while in ovarian cancer cells, only in the fucoidan group, antiproliferative effects were detected at molecular level.
