Production of Polystyrene Based Nanofibers by Electrospinning and Applications of its for the Preparation of Biofunctional Surface
Project no. 20482 · Status: Kapanmış · 2019
Summary
Diagnostic techniques based on biomolecules; The food/beverage industry has application potential that can be used in many fields of study, such as disease diagnosis, bioprocess imaging and environmental pollutant imaging. The success of immobilization on the conductive surfaces of biomolecules plays a key role in increasing the stabilization, sensitivity and selectivity of biomolecules to be used in bioassay systems. Nanofibers are a good candidate for immobilization of biomolecules due to many advantages such as morphology and pore size. Montmorillonite clay (Mt) was modified with PAMAM G3 and added to the PS solution, and nanofibers were obtained by the electrospinning method. The nanofibres obtained by testing %PS and %Mt- PAMAM G3 ratios were characterized by SEM. Anti-Serum Amyloid A antibody was then conjugated to the surface with covalent bonds using EDC-NHS chemistry to the GCE surface. Finally, the selective surface obtained was used for the electrochemical determination of Serum Amyloid A levels. The linear range of PS/Mt-PAMAM/Anti-SAA biosensor is between 1 ng/ml - 200 ng/ml. The detection limit of the PS/Mt-PAMAM/ Anti-SAA biosensor is 0.580 ng/ml SAA. The applicability of PS/Mt-PAMAM/Anti-SAA biosensor was investigated by taking measurements with synthetic saliva and serum containing SAA.
