Investigation of the anti-cancer properties of BOR mineral (disodium pentaborate decahydrate) in ovarian cancer cells.

Project no. 20641 · Status: Kapanmış · 2019

Summary

Ovarian cancer is the deadliest cancer among gynecological malignancies and the 5th most important cause of death among female cancers. The lifetime risk in a woman is reported as 1/54-1/70. Since the disease does not show symptoms in the early period and there is no effective screening method, 70% of the patients are diagnosed in the advanced stage. Recurrence is seen at a rate of 40-50% within 2 years in advanced stage cases in which complete remission is achieved in treatment. The mainstay of treatment is cytoreductive surgery and chemotherapy. Although platinum and taxol-based agents are used in chemotherapy, platinum-resistant cases progress very poorly and an effective treatment approach is unfortunately not found in these cases. Boron mineral, which we have the richest reserves in the world in our country, is among the trace elements taken with food, but it is also used as a food supplement recently. In recent years, molecular studies have focused on targeted therapies in order to increase the effectiveness of chemotherapy and to reduce the effective dose and prevent drug side effects. Although there are many studies in this regard in ovarian cancer, only two agents have been approved (VEGF monoclonal antibody and PARP inhibitor). mTOR signaling pathway; In cancer cells, it involves activation of receptor tyrosine kinases (RTK), which trigger cytoplasmic kinases (such as serine/tyrosine kinases) involved in signal transduction. Thanks to this activation, the mTOR signaling pathway ensures the return of cancer cells. There are studies showing the anticancer properties of many boron compounds. However, according to the current literature, there is no study showing the effects of DPD on ovarian cancer. In our study, we investigated the effects of DPD on the cytotoxic, apoptotic and mTOR pathway gene expressions on the SKOV3 cell line. In cytotoxicity studies, IC50 values were found to be 6.7 mM at 24th hour, 5.6 mM at 48th hour, and 5.2mM at 72nd hour as a result of DPD application to SKOV3 cells in a concentration range of 1.5-20 mM. In apoptosis experiments, when compared to the control group, apoptosis increased 2.6 times, according to the Annexin V staining method, 4.5 times compared to the JC1 method, when compared to the control group, in SKOV3 cells treated with 5.6 mM DPD substance for 48 hours. 84 gene expression changes associated with mTOR mechanisms were detected by real-time qRT–PCR. According to the results of the gene expression analysis, significant expression changes were detected in 54 genes and contributed to the literature as a guide to further molecular studies.
