Summary

Colon cancer is the third leading cause of death from cancer in the world. It is the most common type of cancer among the cancer types in the gastrointestinal tract. When we look at the general treatment options of the disease, radiotherapy, chemotherapy and surgical intervention are the leading ones. The type of treatment is personal and determined according to the stage of the cancer.

The most basic chemotherapeutic agent used in colon cancer is 5-Fluorouracil (5-FU), but the overall survival rate in the use of this drug is low. 5-FU shows its effect through the cell cycle. This agent inhibits DNA replication by inhibiting the cell cycle in S phase. In addition, this chemotherapeutic drug is an antimetabolite in the fluoropyrimidine form that induces cell death and can be administered alone or in combination with other chemotherapeutic agents.

Propolis is a natural ingredient that has been known for centuries and has been studied for years. As a result of the studies, it has been determined that this natural component has properties such as anti-inflammatory, anti-bacterial, anti-oxidant. It has been shown that propolis contains more than 300 compounds such as aromatic acids, benzoic acid and its derivatives, flavonoids, but it is known that the majority of its content consists of flavonoids. Solvents such as methanol, ethanol, water and chloroform are generally used for the extract of propolis, but the most efficient result was obtained from the extract using ethanol. In addition, it is known that this form of propolis is mostly used in studies conducted today.

The overall survival rate is low when chemotherapeutic agents are administered alone. It is applied in combination with natural ingredients to increase this rate. There is no combination study of 5-FU and Turkish propolis in the literature. We evaluated the cytotoxic and apoptotic effect of combination administration in HCT-116 colon cancer cells, aiming to increase the overall survival rate. Cells without active substance are the control group. In addition, we used the WI38 healthy fibroblast cell line to evaluate the effect of the combination in healthy cells.

With the proliferation experiment, which is the first step of our research, the appropriate amount of cells per well was determined to be used in further experiments. Then, the cytotoxic effect of single applications for both agents was determined by MTT method. Combination application was made according to the determined IC50 values. Its apoptotic effect was determined by the Annexin V-FITC method.

The IC50 of propolis for the HCT116 cell line was 407.7 μg/mL (72h); The IC50 value of 5-FU was determined as 9.5 μM (72nd hour).

In the light of all these findings, the effect of combination application of propolis and 5FU active substances on the viability of colon cancer cells in vitro has been demonstrated and it is aimed to initiate further phase experiments to create new treatment options.
