Endüstriyel Atıksulardan Elde Edilen Bazı C. testosteroni suşlarının Fitalat, İzofitalat ve Terefitalat İzomerlerini Parçalama Kapasitelerinin Araştırılması.

Project no. 593 · Status: Kapanmış · 2016

Summary

Within the scope of this research project, three different phthalic acid isomers (Phthalic acid (PA), Terephthalic acid (TPA) and Isophthalic acid (IPA)) were examined with eleven C. testosteroni isolates obtained from an aeration pool of an industrial wastewater treatment plant where plastic raw material waste has been intensively piled up. The degradation activities of eleven C. testosteroni isolates on three phthalate isomers were determined analytically by HLPC instrument. All three chemicals were studied at an optimum concentration as 100 ppm for the observation of degradation activities. Degradation conditions were carried out at 30 ° C in a 200 rpm shaking incubator. Activity isolates for all three chemicals were identified separately. Subsequently isolates with the best activity were selected from C. testosteroni isolates, which are capable of degrading PA, TPA and IPA. Two C. testosteroni strains (C10 and C11) were found to act faster and efficiently among other 9 isolates. At the onset of the study, degradation kinetics and expression levels with the presence of potential genes that might be responsible for degradation of them were investigated for isolate C10 and C11. It was determined that the isolate C10 degrades IPA and the isolate C11 degrades PA and TPA efficiently within the light of obtained data. C10 degrades IFA within 7 hours, while C11 degrades PA and TPA within 6 hours. Among the genes thought to be effective in PA, TPA and IPA degradation; ophA gene for PA, tphA2 and tphA3 genes for TPA, and iphA gene for IPA were detected. In addition, the pmdA and pmdB genes show that these chemicals are broken down to carbon dioxide and water in the metabolic pathway and this means that chemicals are completely metabolised by these isolates. The presence of these genes has also been determined. Alterations in expression levels were detected in all genes other than the iphA gene, with different expression at different times for each chemical at the gene expression level.
