Production Of Fibrin Sealant-Based Biological Contact Lenses Functionalized For Drug Release
Project no. 32672 · Status: Kapanmış (Yayın bekleniyor) · 2025
Summary

Frequent administration of antibiotic eye drops for the treatment of corneal infections may reduce patient adherence and increase treatment burden. In this study, a personalized fibrin sealant-based biological contact lens was developed for controlled vancomycin delivery, and its in vitro feasibility was evaluated.

Ocular surface molds obtained from postmortem small-ruminant eyes collected from a slaughterhouse were digitized using optical three-dimensional (3D) scanning, processed with computer-aided design software, and used to fabricate personalized molds by stereolithography (STL). Vancomycin-loaded fibrin sealant-based biological contact lenses were subsequently prepared using these molds. The structural integrity of the system and its vancomycin release profile were evaluated over 72 h in distilled water and phosphate-buffered saline (PBS).

The developed manufacturing approach enabled the successful fabrication of biological contact lenses conforming to ocular surface anatomy, while the lenses maintained their overall structural integrity throughout the evaluation period. Vancomycin exhibited a biphasic release profile in both media. Cumulative release during the first 24 h reached 52.40% in distilled water and 50% in PBS, increasing to 59.22% and 77%, respectively, at 72 h. These findings indicate an initial rapid release phase followed by a slower, more controlled release phase.

In conclusion, personalized fibrin sealant-based biological contact lenses can be fabricated using three-dimensional modeling and printing technologies and can provide vancomycin release for at least 72 h. This approach provides a promising basis for further formulation optimization and in vivo evaluation of controlled ocular antibiotic delivery systems.

Keywords: Infectious keratitis; fibrin sealant; controlled release; biological contact lens.
