Expression and purification of Toxoplasma gondii GRA1 and ROP6 recombinant proteins in HEK293F cells in different media
Project no. 29652 · Status: Kapanmış (Yayın bekleniyor) · 2023
Summary

T. gondii, an obligate intracellular protozoan parasite, has the potential to infect all mammals. There is no effective drug at the desired level in the treatment of toxoplasmosis, which causes serious clinical pictures, deaths and economic losses in both humans and farm animals. For these reasons, the need for a safe and effective vaccine against T. gondii that can be used in both humans and animals, and therefore research on this subject is continuing intensively. Vaccine research against T. gondii has accelerated in recent years with the discovery of different expression systems and the introduction of new adjuvants, as well as developing bioinformatics programmes and in vitro screening tests. In this project, GRA1 protein, which was determined to be antigenic in previous studies of our research group, was successfully produced in suspended mammalian HEK293 cells and purified for the first time. Recombinant ROp6 protein has not been expressed abundantly. Although the amount of recombinant protein produced is less in quantity, more costly and labour intensive compared to bacterial systems, the ability of proteins obtained with HEK293, one of the human mammalian cell lines, to express proteins with post translational modifications is very important in terms of increasing the antigenic properties of vaccine candidate proteins. With this research, the expression process of recombinant GRA1 protein in mammalian cells was defined for the first time and it was aimed to pave the way for new vaccine designs in the future.
