Design of Bioluminescent Based Microbial Biosensor as a Pesticide Detector in Sustainable Agriculture
Project no. 25241 · Status: Kapanmış (Yayın bekleniyor) · 2022
Summary

Pesticides are widely used in agricultural production to increase yield; however, they may accumulate in the environment and cause adverse effects on ecosystem and human health. Therefore, the rapid, sensitive and reliable detection of pesticide residues in environmental matrices is of great importance. Although conventional analytical methods provide high accuracy, they can be time-consuming and costly. In contrast, microbial biosensors sensitive to toxic compounds offer alternative approaches that enable rapid and on-site detection of environmental pollutants.

In this context, the aim of this study was to determine the bioluminescence-based responses to the pesticides cyprodinil, chlorpropham and chlorpyrifos, which are commonly used in agricultural areas, and to evaluate the applicability of these responses in biosensor applications. Within this scope, bioluminescent bacteria were isolated from marine samples and the obtained isolates were molecularly identified based on the 16S rDNA gene region and determined to belong to the genus Vibrio. Among the isolated bioluminescent bacteria, the strain showing the most pronounced bioluminescence response, Vibrio sp. SL-2, was selected and the experimental studies were continued with this strain. The selected strain was exposed to different concentrations (100 ppm, 10 ppm, 1 ppm and 0.01 ppm) of cyprodinil, chlorpropham and chlorpyrifos, and the resulting bioluminescence responses were analyzed using a digital imaging system in a dark environment and a luminometer device. The results showed that the SL-2 strain exhibited distinct and dose-dependent bioluminescence responses to each of the tested pesticides, particularly at concentrations of 100ppm and 10 ppm. These findings indicate that this strain may serve as a potential biological indicator for biosensor-based applications that can be used in environmental monitoring studies due to its sensitive and measurable bioluminescence responses to pesticides.
