Development of Effective Film Systems for the Treatment of Cold Sores Caused by Herpes Simplex Virus
Project no. 29726 · Status: Kapanmış (Yayın bekleniyor) · 2023
Summary

This project aimed to develop film systems to support the alleviation of symptoms of herpes simplex virus-induced cold sores. Therefore, polymeric film formulations were prepared using chitosan, sodium alginate, and pullulan polymers to create a mechanical barrier in the lesion area and increase patient compliance. Additionally, ursolic acid, which has antioxidant and antimicrobial effects, was added to the formulations. The prepared films were characterized in terms of macroscopic properties, thickness, weight, pH, moisture content, dispersion time, water retention capacity, mucoadhesion, mechanical strength, and microscopic morphology. Furthermore, sterilization, microbiological control, antimicrobial efficacy, antioxidant capacity, biocompatibility, and stability studies were conducted.

The results showed that pullulan-based film formulations exhibited more favorable properties compared to other formulations in terms of appearance, flexibility, homogeneity, mucoadhesion, and stability under normal storage conditions. Ursolic acid, possessing antimicrobial and antioxidant activity, was added to the formulation. Antioxidant activity was observed in formulations containing ursolic acid, but no significant antimicrobial activity was detected under the tested conditions. However, microbial quality studies showed that ursolic acid-loaded formulations were microbiologically protected. The cytotoxicity of the products was evaluated in two different studies. In addition, studies conducted within the scope of biocompatibility showed that the selected pullulan-based formulation was suitable in terms of dermal irritation and sensitization. In conclusion, the developed film systems are promising for an innovative medical device approach in the management of herpes lesions. Herein, an innovative product has been developed to alleviate herpes symptoms, prevent the spread of herpes to surrounding tissues, and contribute to the healing process of lesions.
