INVESTIGATION ON EFFECT OF CYTOKININ SIGNAL COMPONENTS WHICH NEGATIVELY REGULATE DROUGHT RESPONSE ON REACTIVE OXYGEN SPECIES (ROS) IN ARABIDOPSIS

Project no. 423 · Status: Kapanmış · 2017

Summary

Plants are exposed to many stresses in their environment. Drought stress is the most harmful of these stresses. This study is about investigating the effect of ARR1/10/12, a cytokine signaling pathway component, on the regulation of reactive oxygen species (ROS) under drought stress. As plant material Arabidopsis thaliana Col. wild type and Arabidopsis thaliana ARR1/10/12 knock-downed triple mutant had been used. After the plants were grown in the soil environment, the physiological and biochemical parameters were determined by harvesting in three periods (vegetative, flowering and rewatering period). As physiological parameters; body wet weights, relative water contents, stoma openings, stoma counts per unit area, and leaf water losses were analyzed. As biochemical parameters; lipid peroxidation, H2O2 content, antioxidant enzymes / isoenzymes activity (catalase (CAT), superoxide disutase (SOD), peroxidase (POX), cell wall POX (CWPOX), ascorbate peroxidase (APOX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), NADPH oxidase (NOX)) and nonenzymatic antioxidants (ascorbate (ASC), GSSG and GSH) had been measured
