EFFECTS OF BORON MINERALS ON APOPTOSIS AND GENE EXPRESSION IN OVCAR-3 OVER CANCER CELLS
Project no. 23178 · Status: Kapanmış (Yayın bekleniyor) · 2021
Summary

Ovarian cancer is the deadliest cancer among gynecological malignancies and the 5th most important cause of death among female cancers. The lifetime risk in a woman is reported as 1/54-1/70. Since the disease does not show symptoms in the early period and there is no effective screening method, 70% of the patients are diagnosed in the advanced stage. Recurrence is seen at a rate of 40-50% within 2 years in advanced stage cases in which complete remission is achieved in treatment. The mainstay of treatment is cytoreductive surgery and chemotherapy. Although platinum and taxol-based agents are used in chemotherapy, platinum-resistant cases progress very poorly and an effective treatment approach is unfortunately not found in these cases.

Boron mineral, which we have the richest reserves in the world in our country, is among the trace elements taken with food, but it is also used as a food supplement recently. When we look at the field of health, it is seen that boron compounds are used in osteoporosis and rheumatoid arthritis treatments, burn treatments, wound healing, antiseptic lens solutions, ointments, mouthwashes and eye drops.

In our study, we investigated the effects of DPD on the cytotoxic, apoptotic and mTOR pathway gene expressions on the OVCAR-3 cell line. In cytotoxicity studies, IC50 values were found to be 3,2 mM at 24th hour, 1.4 mM at 48th hour, and 1,5 mM at 72nd hour as a result of DPD application to OVCAR-3 cells in a concentration range of 0.01-5 mM.

In apoptosis experiments, it was shown that apoptosis increased 2.2 times according to the Annexin V staining method when compared to the control group in OVCAR-3 cells, which were applied 1.4 mM DPD substance for 48 hours.

Expression changes of 84 genes related to mTOR mechanisms were detected by real-time qRT–PCR. According to the results of the gene expression analysis, significant expression changes were detected in 27 genes and contributed to the literature as a guide to further molecular studies.

Keywords; Ovarian cancer, Boron, cell line, metastasis, proliferation
