Isolation, Identification and Investigation of Acetylcholinesterase Inhibition Activities of Galanthus elwesii and Endemic Galanthus trojanus Endophytes Distributed in the Aegean Region
Project no. 22795 · Status: Kapanmış (Yayın bekleniyor) · 2021
Summary

Amaryllidaceae family plants have been used for medicinal purposes for centuries thanks to the alkaloids they contain. Although Amaryllidaceae alkaloids have many medical effects, they are currently the most effective drugs used in the treatment of Alzheimer's disease (AD) thanks to their acetylcholinesterase (AChE) inhibition activities. In recent years, with the understanding that biosynthesis pathways are shared between plants and their endophytes, the search for new metabolites has turned to endophytic microorganisms. In literature research conducted in this context, it has been observed that the endophytes of plants with AChE inhibition activity also have AChE inhibition activity. For this reason, in the proposed project, Galanthus elwesii Hook, a member of Amaryllidaceae that is rich in bioactive alkaloids and produces galantamine used in the treatment of AD and is distributed in Western and Southern Anatolia in Turkey. f. and Galanthus trojanus A.P., one of the endemic species of Turkey, distributed in Balıkesir, Çanakkale. Davis & N. Özhatay endophytes were selected to be investigated for their AChE inhibitory activities. Within the scope of the project, G. elwesii plants from the Aegean region and G. trojanus plants from the Marmara region will be collected and surface sterilization will be carried out. Sections will be taken from the internal tissues of the plants and transferred to the relevant media for endophyte isolation. Isolated microorganisms will be purified and identified using the polyphasic approach. After the fermentation of endophytes is done, the extraction of alkaloid metabolites will be carried out by liquid-liquid extraction. The AChEI activity of the obtained extracts will be determined by the modified Ellman method, and the active compounds in the extracts showing AChEI activity will be determined by the AChE-based thin layer chromatography (TLC)-autography method.

As a result, the endophytes of G. elwesii and endemic G. trojanus plants, which are an important part of Turkey's biodiversity, will be identified for the first time and introduced to the literature, and the AChE inhibition activities of the endophyte isolates will be determined. If strains showing inhibitory activity are identified, a great potential will be revealed for the production of Galantamine-like/derivative molecules by defining the molecular structures of active compounds through further projects.
