HeLa Servikal Kanser Hücre Hattında JQ1 ve Temozolomide İlaç Kombinasyonunun Sitotoksik ve AntiProliferatif Etkilerinin İncelenmesi

Project no. 23416 · Status: Kapanmış · 2021

Summary

Cervical cancer is the second most common gynecological cancer in women worldwide. In Turkey, it is the third most common cancer in women after breast cancer and corpus uteri cancer. The resistance to chemotherapy seen in cervical cancer cells makes the treatment of advanced cervical cancer difficult. Aldehyde Dehydrogenase (ALDH), a family of enzymes that catalyze the oxidation of toxic aldehydes and other harmful chemical compounds and the high level of ALDH1 expression seen in some cancer cells has been found to play a key role in chemotherapy resistance. The effects of ALDH1 suppression in glioblastoma (GBM) cell line and some other tumor cells were tested and it was observed that the cytotoxic effects of the chemotherapy agent increased. Temozolomide (TMZ), which has similar effects with traditional alkylating agents, is currently used in the treatment of GBM. Studies have shown that TMZ becomes an effective chemotherapeutic agent by suppressing ALDH activity in GBM cell line with the help of Bromodomain Extraterminal motif (BET) inhibitors. This suggests the hypothesis that the same effect can be observed in cervical cancer cells. Although there are studies in the literature where the BET inhibitor is applied on the JQ1 cervical cancer cell line, there is no publication examining the joint effect of BET inhibitor and TMZ on the cervical cancer cell line so that makes our project unique from other research. Therefore, in the project proposal, it is aimed to find the answer to the question of how effective the joint use of TMZ and BET inhibitor JQ1 can be on cervical cancer cells.
