Summary
Influenza caused by influenza virus is a viral disease that is generally acute and has a high transmission rate, is globally widespread, causing epidemics and pandemics. The circulating virus strain of the RNA influenza virus, which has a high rate of antigenic mutations, therefore differs. In this context, the identification and production of the strains to be included in the seasonal flu vaccines, which are re-developed every year, is an important problem and may cause disruptions in meeting the world vaccine needs. Although there are some anti-viral drugs that can be used in the treatment of flu infections, prevention is the best option. The best way to prevent influenza infection is vaccination. Inactive whole virus vaccines are widely used in the world. Every year, the World Health Organization declares influenza vaccine strains for the northern and southern hemispheres. For this purpose, trivalent and quadrivalent vaccines are produced in recent years. The most important vaccine component in the development of inactive vaccines is adjuvant-adjuvant systems. Research on adjuvant systems that have the effect of increasing the protective potential of the vaccine has increased in recent years, and it is important to investigate saponins isolated from our country's endemic plants, which may be an alternative to the natural product-induced QS-21 saponin adjuvant. The aim of this study was to investigate the potential use of Scoposide A, Scoposide B, Elmalienoside A, Balansoide B, Aristatoside B saponins as adjuvants in inactivated virus vaccine formulations against seasonal influenza (H3N2) in vitro. After the adjuvant-antigen formulations were prepared, the immunomodulatory potentials of the formulations were determined by conducting IL-4, IL-6, TNF-?, IL-1? and IFN-? cytokine analysis by ELISA. When ELISA analysis results are examined, in IFN-? concentration results) compared to control DMSO; Scoposide B (p <0.002); Elmalienoside A (p <0.01); Balansoide B (p <0.05) increase was observed. When the IFN-? concentration results of vaccine formulations made with H3N2 antigen are examined; in comparison with H3N2 as control; formulated with H3N2 antigen; Scoposide A (p <0.0001); Scoposide B (p <0.0001); Elmalienoside A (p <0.0001); Balansoide B (p <0.0001); Aristatoside B (p <0.0001) increase was observed. In addition, the formulation prepared with H3N2 and Balansoide B saponin also showed an increase in IL-1? (p = 0.005). It has been determined that, primarily Balansoide B; Scoposide A, Scoposide B, Elmalienoside A, Aristatoside B saponins obtained from the Cephalaria spp. plant species, which endemically grow in our country due to the high antiviral properties of IFN-?, have high potential as adjuvants in seasonal influenza (H3N2) vaccine formulations.
