Project no. 21867 · Status: Kapanmış · 2020

Summary

The plant world is the source of a wide variety of natural compounds with different chemical structures, with a wide spectrum of biological activities, most of which find application in health sciences. Natural compounds have been the primary source of precursors and commercial drugs until recent years. Recent research has shown that 61% of the 877 drugs known worldwide are of natural compound origin. It is thought that there are about 350,000 plant species in the world, one third of which has not yet been discovered. Chemical studies have been carried out on only fractions of a quarter of a million of those discovered.

The Caryophyllaceae family is a herbaceous and flowering group that grows in moderate climates and warm regions, spreading in the Northern hemisphere, especially in the Mediterranean Region. In fact, the term saponin derives from the plant Saponaria officinalis from the family Caryophyllaceae, whose roots have been used as soap since ancient times. Gypsophila paniculata/muralis and Acanthophyllum gypsophiloides are other well-known saponin-containing plants in this family. The use of Dianthus chinensis, Vaccaria segetalis and Silene species from the Caryophyllaceae family as herbal medicine in Asia, especially in Chinese medicine, has been recorded. Saponaria officinalis and Gypsophila species are also used as traditional medicine in Europe.

Considering previous studies and literature data, in this project proposal, Moenchia mantica subsp. The aim of this study was to determine the cytotoxic activities of isolated compounds by isolation, purification and characterization of secondary metabolites from mantica. For this purpose, dried and ground plant materials will be extracted using solvents of different polarity, and the extracts will be obtained sequentially with different fillers by using column chromatography methods (VSK-VLC, open column chromatography, OBSK-MPLC) to obtain pure components. will go his way. Structure determinations of the purified compounds will be performed using extensive 1D- and 2D-NMR techniques. The cytotoxic activities of the isolated pure compounds will be tested by MTT method using A-549 (human lung carcinoma), U87MG (glioblastoma cell line) and normal cell line as control.

Recent studies show that metal-based anti-cancer drugs play a critical role in the prevention of all types of cancer, but their efficacy includes normal cells as well as cancer cells. However, metal N-heterocyclic carbene (NHC) complexes show great potential as they show the potential to selectively destroy cancer cells rather than healthy cells, especially by targeting cancer cells leading to cell death or inhibition of cell growth.

Metal complexes with NHC ligands are widely used in catalytic chemistry. The strong σ-donor properties of NHCs, the strength of metal NHC bonds and their resistance to oxidation cause metal-NHC complexes to have high chemical and thermal stability. It is seen as an emerging research area for the development of In addition, research on the biological potential of metal complexes with NHC ligands is one of the most active areas of bioorganometallic chemistry, as they perfectly match the prerequisites for efficient drug design and rapid optimization.

In the second stage of our project proposal, it is planned to investigate the cyctotoxic activities of NHC-iridium transition metal complexes, whose steric and electronic properties can be easily modified.
