Summary

Polycystic ovary syndrome (PCOS) is a common and complex endocrine disorder observed in women of reproductive age. However, the pathogenesis of PCOS still remains unclear. The aim of this study is to investigate the possible therapeutic effects of ISO-1, a macrophage migration inhibitory factor (MIF) inhibitor, in a letrozole-induced PCOS mouse model from a histological perspective.

In this study, female Balb/c mice were divided into four groups (n=10) and administered 1 mg/kg letrozole by gavage for 21 days to establish a PCOS model. To examine the effects of the MIF inhibitor ISO-1, the animals were treated with 4 mg/kg ISO-1 intraperitoneally for 14 days. At the end of the experiment, ovaries were collected and subjected to histochemical, immunohistochemical, and molecular analyses including RT-PCR and ELISA. The results indicated that MIF inhibition with ISO-1 in PCOS-induced mice affected folliculogenesis and could influence the inflammatory processes associated with PCOS.

In addition to assessing the molecular regulation of MIF expression levels alongside other inflammatory biomarkers, the evaluation of ISO-1’s impact on ovarian morphology in the PCOS mouse model highlights the originality of this study and provides a foundation for future research.

A review of the literature revealed no previous studies investigating the potential therapeutic effects of ISO-1 on the ovary in a PCOS mouse model. This supports the originality and novelty of the findings presented in this study.
