Novel Approaches in Breast Cancer Therapy

Project no. 291 · Status: Kapanmış · 2018

Summary

The possible chemopreventive effect of melatonin was reported to mediated through its oxidative stress modulating and inhibitory effects on cell proliferation pathways and aromatase by binding to the melatonin receptor. Melatonin receptor agonists, agomelatine (approved by the EMA-European Medicines Agency for the treatment of depression indication) and ramelteon (approved by the FDA-US Food and Drug Administration for the treatment of sleep disorders) are molecules with high affinity to melatonin receptors. However, in the literature, there is no information about the cytotoxic effects of these molecules on breast cancer cells. Based on this information, within the scope of the project, the cytotoxic and antiproliferative effects of melatonin receptor agonists on human breast cancer cells were investigated by in vitro MTT, LDH and colony formation experiments. According to the findings, agomelatine and ramelteon showed cytotoxicity in breast cancer cells at 10-4 M concentration without affecting normal breast epithelial cell viability. By evaulating melatonin receptor agonists for their cytotoxicities, the first step to find more effective candidates for the treatment of breast cancer and/or the prevention from breast cancer in high risk carrier individuals was achieved.
