The Phytochemical and semisynthetic studies on Scabiosa pseudograminifolia

Project no. 22215 · Status: Kapanmış · 2020

Summary

The Scabiosa, belonging to the family Caprifoliaceae, has recently been highlighted in numerous studies as a rich source of biologically active compounds such as saponins and iridoids, with significant potential for biological activity. In this thesis, the detailed phytochemical investigation of Scabiosa pseudograminifolia was conducted for the first time, leading to the isolation and structural elucidation of four new saponins (1-4), ten known saponins, a triterpene aglycone (oleanolic acid), and seven iridoid glycosides, three of which are bis-iridoids. The effects of all the isolated pure compounds on macrophage polarization and their cytotoxicities on the healthy CCD-34Lu cell line were evaluated. Notably, saponins were found to modulate immune responses by influencing macrophage polarization between M1 (pro-inflammatory) and M2 (anti-inflammatory) phenotypes, indicating their potential as adjuvant drugs in cancer therapy. Additionally, oleanolic acid, which was obtained as a major compound, was used as a starting material for the synthesis of new amide derivatives, and the cytotoxic activities of these derivatives were assessed on various cell lines. These findings not only define the chemical composition of S. pseudograminifolia but also provide valuable insights into the biological activities of its compounds and derivatives, paving the way for their potential future medical and industrial applications
