The Effect of Regular Aerobic Exercise on Hepcidin and Hematological Parameters in Football Referees and The Role of TMPRSS6 RS855791 Polymorphism

Project no. 683 · Status: Kapanmış · 2018

Summary

Introduction and aims: Hepcidine is a main hormone that controls iron metabolism. The effects of rs 855791 polymorphism (TMRP) of TMPRSS6 gene associated with hepcidine and the effects of exercise on the level of hepcidine and main hematological parameters is unclear. The aim of this study was to investigate the effect of aerobic exercise on hepcidine and some hematological parameters and the role of TMRP. Method: 46 healthy trained male soccer referee (23.3±2.8 years) and 43 sedantary (23.2±3.3 years) participated to this study. The yo-yo intermittent (maximal) exercise (level 2) test was performed as an endurance criterion. Using postprandial of venous blood samples; hepcidine and oxidized low density lipoprotein (oxLDL) levels (by ELISA method), total antioxidant/oxidant status (TAS and TOS), hemogram, ferritin, iron and iron binding capacity, thiobarbituric acid reactive substances (TBARS) were determined by standard methods; (TMRP) was determined by restriction fragment length polymorphism analysis after polymerase chain reaction from genomic DNAs isolated from periferic blood samples. Results: The aerobic capacity (yo-yo test value) of all athletes including genotype groups was significantly greater than that of the control groups. The iron and mean erythrocyte volume (MCV) (p = 0.000) values of athlletes were significantly lower than control (p = 0.018). TAS values of athlletes were significantly greater (p = 0,000) than control, but the value of OxLdL (p = 0,011) ,OSI (TOS / TAS ratio) (p=0,008), MCV ( p = 0.045) and iron (p = 0.000) values were lower than the control. No significant difference was found between the groups for other oxidative stress parameters (p > 0.05). At athletes (n=46); MCV values of T carrier group (Tc =TT + TC) were significantly lower than the controls (p = 0.035); whereas the number of erythrocytes (p = 0.004) and TAS (p = 0.001) (depending on TMRP) was greater than the control (n=30). No significant difference was found between the groups for hepcidin (p > 0.05). At the athletic groups; TBARS values of Tc group were significantly greater than those of the homozygous CC group (p = 0.00). At control genotype groups, iron, erythrocyte values of Tc group were significantly greater than that of the homozygous CC group (p < 0.05). Conclusion: These findings show that, the regular aerobic exercise; a) Increased the capacity of aerobic and antioxidant at athletic group. b) Increased the number of erythrocytes in the Tc group depending on the TMRP (without a significant change at the hepcidine levels). c) Decreased athletic groups’s iron levels and didn’t change hepcidin levels. In the differences found between each genotype groups of the control and the athletic groups may be role of the TMRP. It is recommended that the athleths should take iron-rich foods.
