Summary

Lung cancer is a common type of cancer and the leading cause of cancer-related death. Cisplatin is a chemotherapeutic agent widely used in the treatment of many cancer types, including lung cancer. It is known that ~60% of patients develop resistance to cisplatin, which is used in the standard treatment of non-small cell lung cancer (NSCLC), over time. Therefore, it is necessary to elucidate the mechanism of cisplatin resistance and identify new molecular targets.

In our team's Priority Area Project No. TOA-2021-22673 titled ‘Identification of Genetic Components Affecting Cisplatin Resistance in Non-Small Cell Lung Cancer Calu-1 Cell Line by Whole Genome CRISPR/Cas9-based Genetic Screening Method’, we identified two new candidate genes responsible for cisplatin resistance. These are GPR89A (G-Protein Coupled Receptor 89A) and PUF60 (Poly(U) binding splicing factor 60) genes, and we confirmed that they are responsible for cisplatin resistance after in vitro validation studies.

Our aim in this proposed project is to demonstrate the effects of GPR89A and PUF60 genes, whose effects we have determined in in vitro cell culture studies, in an in vivo mouse xenograft lung cancer model.
