Summary

Prostate cancer is the second most common cause of cancer-related deaths worldwide in men, following lung cancer. In prostate cancer cases with intermediate risk,short-term hormone suppression therapy combined with radiotherapy, and in high-risk cases ,longer-term hormone treatments are recommended by all international treatment guidelines.Recently, new studies have emerged focusing on increasing the effectiveness of hormone suppression therapies. These new approaches primarily concentrate on androgen receptor inhibition. The discovery that androgen receptors send signals for tumor cell growth has made the concurrent use of these trearments with ionizing radiation a significant area of research. In our study,we aim to investigate the effects of androgen receptor blockade on cell death and changes in the cycle in prostate cancer cell lines.

In our study ,prostate cancer cell lines were used and these cell lines were first exposed to ionizing radiation in order to determine the lethal dose. After this step, the cells were divided into three different treatment groups;ionizing radiation + olaparib, ionizing radiation + enzalutamide and ionizing radiation + abiraterone acetate. The cell lines were then compared in terms of cell death and changes in the cell cycle. In our study, the effects of androgen receptor blockade with ionizing radiation,as well as the differences between these treatments, were evaluated. The results revealed that the simultaneous use of androgen receptor blockade with ionizing radiation increased cell death, independent of androgen sensitivity. However,in the comparisons made, enzalutamide was identified as the androgen receptor blocker that caused the most cell death, even at the lowest dose. Adding olaparib caused cell death with all androgen reseptor blocers.
